ABSTRACT

In the Myrtaceae family, the species Eugenia involucrata DC., popularly known as "cerejeira-do-mato", is traditionally used for the antidiarrheal and digestive action of its leaves. However, no studies were found in the literature regarding its antimicrobial and antioxidant potential. In this context, the objective of the present study was to determine the chemical composition by gas chromatography coupled to mass spectrometry (GC-MS) to evaluate the antimicrobial activity by the broth microdilution technique and the antioxidant activity by the 2,2-diphenyl-1-picryl-hydrazila (DPPH) method of the essential oil of E. involucrataleaves. GC-MS identified 28 compounds, all sesquiterpenes, corresponding to 89.41% of the essential oil. The antimicrobial activity of the essential oil was observed for all Gram-positive bacteria tested (Staplylococcus epidermidis, Enterococcus faecalis,Bacillus subtilis and Staplylococcus aureus) and for yeast Candida albicans. The essential oil presented a reduction capacity of DPPH up to 66.81%, evidencing its antioxidant potential. It is suggested that the antimicrobial and antioxidant action of E. involucrata essential oil is related to the presence of the major compounds, elixene (26.53%), ß-caryophyllene (13.16%), α-copaene (8.41%) and germacrene D (7.17%).


Na família Myrtaceae, a espécieEugenia involucrata DC. popularmente denominada "cerejeira-do-mato" é conhecida tradicionalmente pela ação antidiarreica e digestiva de suas folhas. Contudo, na literatura não foram encontrados trabalhos referentes ao seu potencial antimicrobiano e antioxidante. Neste contexto, o objetivo do presente estudo foi determinar a composição química por cromatografia gasosa acoplada a espectrometria de massas (CG-EM) e avaliar a atividade antimicrobiana pela técnica de microdiluição em caldo e a atividade antioxidante pelo método do 2,2-difenil-1-picril-hidrazila (DPPH) do óleo essencial das folhas de E. involucrata. A CG-EM identificou 28 compostos, todos sesquiterpenos, correspondendo a 89,41% do óleo essencial. A atividade antimicrobiana do óleo essencial foi observada para todas as bactérias Gram-positivas testadas (Staplylococcus epidermidis, Enterococcus faecalis,Bacillus subtilise Staplylococcus aureus) e para a levedura Candida albicans. O óleo essencial apresentou capacidade redutora de radicais DPPH de até 66,81%, evidenciando sua potencialidade antioxidante. Sugere-se que a ação antimicrobiana e antioxidante do óleo essencial de E. involucrata esteja relacionada à presença dos compostos majoritários, elixeno (26,53%), ß-cariofileno (13,16%), -copaeno (8,41%) e germacreno D (7,17%).

ABSTRACT: The interest in the use of green tea (Camellia sinensis) (L.) Kuntze (Theaceae) products have increased in the last few years due to its medicinal properties. In the present study, we proposed that headspace solid phase microextraction, in combination with gas chromatography coupled to mass spectrometric could be an efficient method to assess the volatile compounds and to ensure the quality control of C. sinensis. We have also compared the anatomical leaf of different commercial green tea samples, analyzed the information in their labels and determined the presence of foreign materials, moisture content, total ashes and microorganisms. Among the 30 analyzed samples, six were identified as C. sinensis according to the anatomical study; 83.4% were not in accordance with the specified limits for foreign matter and 6.6% for moisture content. All samples presented total ashes in accordance with the current legislation. The analysis indicated the presence of several different volatile organic compounds, being terpenes, hydrocarbons and alcohols the major ones. Microbiological analysis showed that 13.3% and 16.6% of the samples exceeded the limits for counts of molds and yeasts, and for mesophilic bacteria, respectively. 73.3% presented E. coli above the established sanitary limits.


RESUMO: O interesse no chá verde comercial (Camellia sinensis) (L.) Kuntze (Theaceae) tem aumentado nos últimos anos devido suas propriedades medicinais. No presente estudo utilizamos a microextração em fase sólida, em combinação com a cromatografia gasosa acoplada com espectrometria de massas, como um eficiente método de caracterização de compostos voláteis e efetuamos o controle de qualidade de C. sinensis. Comparamos também a anatomia das folhas de diferentes amostras comerciais de chá verde, as informações de seus rótulos e determinamos a presença de matérias estranhas, de microrganismos, o teor de umidade e de cinzas totais. Entre as trinta amostras analisadas, seis foram identificadas como C. sinensis de acordo com o estudo anatômico; 83,4% não estavam de acordo com os limites especificados para matérias estranhas e 6,6% para o teor de umidade. Todas as amostras apresentaram cinzas totais de acordo com a legislação vigente. As análises evidenciaram diversos compostos orgânicos voláteis distintos, sendo majoritários terpenos, hidrocarbonetos e álcoois. As análises microbiológicas mostraram que 13,3 e 16,6% das amostras excedeu os limites de contagem para bolores e leveduras, e para bactérias mesófilas, respectivamente. Por fim, 73,3% das amostras apresentou Escherichia coli acima dos limites sanitários estabelecidos.

Essential oils have aroused interest in the industrial sector due to the multitude of potential applications, especially with respect their antimicrobial and antioxidant properties, among others. The objective of this study was to determine the chemical composition of the essential oil from the leaves of Myrcia oblongata DC by gas chromatography coupled to mass spectrometry (GC-MS). To evaluate the antioxidant potential of the oil by using the free radical capture method with 2,2-diphenyl-1-picryl hydrazyl (DPPH); to test the oil antimicrobial activity using the broth microdilution method; and to evaluate the repellency and fumigant potential of the oil on Dermanyssus gallinae (Degeer, 1778). The GC-MS analysis resulted in the identification of 30 oil constituents, with the bulk of the composition identified as caryophyllene oxide (22.03%) and trans-verbenol (11.94%). The oil presented moderate antioxidant activity compared to the synthetic antioxidant 2,6-di-tert-butyl-4-hydroxytoluene (BHT). Antimicrobial activity of the essential oil showed an inhibitory activity on Gram-positive bacteria, Enterococcus faecalis, Staphylococcus aureus, Bacillus subtilis and Staphylococcus epidermidis and for the yeast Candida albicans, and showed no activity against Gram-negative bacteria. All concentrations of the essential oil used in the fumigation test on D. gallinae resulted in mortality below 20%. For the repellency test, significant potential was observed for the concentration of 10%.


Os óleos essenciais tem despertado interesse no setor industrial por apresentarem multiplicidade de aplicações, destacando-se por suas propriedades antimicrobianas, antioxidantes dentre outras. Diante disto, o objetivo deste estudo foi determinar a composição química do óleo essencial das folhas de Myrcia oblongata DC pelo método de cromatografia gasosa acoplada à espectrometria de massas (CG-EM), avaliar o potencial antioxidante do óleo pelo método de captura do radical livre 2,2- difenil-1-picril hidrazil (DPPH), testar a atividade antimicrobiana pelo método de microdiluição em caldo, bem como avaliar a repelência e potencial fumigante deste óleo sobre Dermanyssus gallinae (Degeer, 1778). As análises de CG-EM resultaram na identificação de 30 constituintes, sendo majoritários o óxido de cariofileno (22.03%) e o trans-verbenol (11.94%). O óleo apresentou atividade antioxidante moderada quando comparada ao antioxidante sintético 2,6-di-tert-butil-4-hidroxitolueno (BHT). Em relação à atividade antimicrobiana, o óleo essencial demonstrou atividade inibitória para as bactérias Gram-positivas, Enterococcus faecalis, Staphylococcus aureus, Bacillus subtilis e Staphylococcus epidermidis, e para levedura Candida albicans e não apresentou atividade para bactérias Gram negativas. Todas as concentrações do óleo essencial testadas no teste de fumigação sobre D. gallinae apresentaram mortalidade inferior a 20%. Para o teste de repelência, observou-se um potencial significativo para a concentração de 10%.

ABSTRACT

ABSTRACT: Essential oils are the most important compounds produced during secondary metabolism in aromatic plants. Essential oils are volatile, have characteristic odor and are used as defensive agents by plants. In pepper, it is possible to say that essential oils are the “flavor fingerprint” of each species. In the present article, eight species of pepper were studied in order to extract their essential oils and oleoresins, test their antibacterial and antifungal activities and also to identify the compounds present in the most bioactive samples. Results demonstrated that two essential oils [Pimenta dioica (L.) Merr. and Schinus terebinthifolius] and three oleoresins (Schinus terebinthifolius and Piper nigrum white and black) recorded significant antimicrobial activity. These active essential oils and oleoresins are interesting for use in biotechnological processes employed in food, pharmaceutical and other industries.


RESUMO: Os óleos essenciais são os compostos mais importantes produzidos durante o metabolismo secundário de plantas aromáticas. No caso das pimentas, é possível afirmar que os óleos essenciais são a “impressão digital do sabor” de cada espécie. No presente artigo, oito espécies de pimenta foram estudadas a fim de extrair seus óleos essenciais e oleoresinas, testar a sua atividade antibacteriana e antifúngica, bem como, identificar os compostos presentes nas amostras mais ativas. Os resultados obtidos mostram que dois óleos essenciais (Pimenta dioica e Schinus terebinthifolius) e três oleoresinas (Schinus terebinthifolius e Piper nigrum branco e preto) apresentaram atividade antimicrobiana significativa. Esses óleos essenciais e oleoresinas bioativos mostram-se interessantes para a utilização em processos biotecnológicos empregados na indústria alimentar, farmacêutica e outras indústrias.

ABSTRACT

INTRODUCCIÓN: diferentes extractos de Terminalia Catappa Linn. (Combretaceae) han demostrado de forma internacional, propiedades farmacológicas beneficiosas para la salud humana. Estas propiedades han sido atribuidas en lo fundamental a los polifenoles y glicósidos, encontrados en hojas, corteza y frutos. En Cuba esta especie es catalogada como una planta invasora y existen pocas investigaciones sobre su composición química y estudios farmacológicos. OBJETIVOS: identificar y cuantificar los ácidos polifenólicos presentes en el extracto metanólico de las hojas de T. catappa utilizándose la cromatografía de gases acoplada a espectrometría de masas. MÉTODOS: las hojas amarillo-rojizas fueron secadas, molidas, desgrasadas con hexano y y con posterioridad extraídas con metanol en un baño ultrasónico. El extracto se filtró y el disolvente se eliminó al vacío. El extracto seco se hidrolizó con ácido clorhídrico y se extrajo con acetato de etilo. Se determinó el rendimiento de extracción, las características organolépticas y los polifenoles totales mediante el método de Follin-Ciocalteu. La composición química del extracto hidrolizado se llevó a cabo por cromatografía de gases acoplada a espectrometría de masas, previa formación de los derivados trimetilsilil. RESULTADOS: se obtuvo un líquido de color pardo rojizo oscuro de olor característico. El contenido total de polifenoles fue 184,6 (mg Pirogalol/100 g Extracto). Se detectaron 37 compuestos por cromatografía de gases acoplada a espectrometría de masas en el extracto metanólico hidrolizado. Este extracto está compuesto de manera general por ácidos polifenólicos como el ácido gálico; ácido vanílico; ácido 3,4-dihidroxibenzoico; ácido 2,5-dihidroxi-benzoico y ácido 4- hidroxibenzoico. También se detectaron otros compuestos con elevados contenidos como ácido elágico y ácido levulínico. CONCLUSIONES: el extracto metanólico de hojas de T. catappa que crece en Cuba mostró un elevado contenido de ácidos polifenólicos, donde los ácidos gálico y elágico fueron los mayoritarios. La presencia de estos compuestos pudiera justificar las propiedades medicinales atribuidas a esta especie, a la vez que servirían de base para continuar con futuras pruebas farmacológicas que avalen sus usos con fines farmacéuticos.


INTRODUCTION: Different extracts of Terminalia Catappa Linn. (Combretaceae) internationally have shown pharmacological properties beneficial to human health. These properties have been largely attributed to polyphenols and their glycosides found in the leaves, bark and fruits. In Cuba this species is listed as an invasive plant and there is limited research on its chemical composition and pharmacological studies. OBJECTIVES: To identify and quantify the polyphenolic acids that could be present in the methanol extract of Terminalia catappa leaves using gas chromatography-mass spectrometry. METHODS: The yellow-red leaves were dried, ground, defatted with hexane and then extracted with methanol in an ultrasonic bath. The extract was filtered and the solvent removed under vacuum. The dry extract was hydrolyzed with hydrochloric acid and extracted with ethyl acetate. The extraction yield, the organoleptic characteristics and the total polyphenols by Follin-Ciocalteu method were determined. The chemical composition of the hydrolyzed extract was performed by gas chromatography-mass spectrometry after formation of trimethylsilyl derivatives. RESULTS: A dark reddish brown liquid with a characteristic odor was obtained. The total polyphenol content was 184.6 (mg Pyrogallol/100g extract). By mean of gas chromatography-mass spectrometry a total of 37 compounds were detected in the hydrolyzed methanol extract. This extract consists mainly of polyphenolic acids such as gallic acid; vanillic acid; 3,4-dihydroxybenzoic acid; 2,5-dihydroxybenzoic acid and 4- hydroxybenzoic acid. Other compounds with high content as ellagic acid and levulinic acid were also detected. CONCLUSIONS: The methanolic extract obtained from the leaves of Terminalia catappa growing in Cuba showed a high content of polyphenolic acids where gallic acid and ellagic predominated. The presence of these compounds could justify the medicinal properties attributed to this species, while providing the basis for further future pharmacological evidence to support its use for pharmaceutical purposes.

ABSTRACT

Smallanthus is a genus of flowering plants in the Asteraceae family, which has about 24 species, ranging mostly from southern Mexico and Central America to the Andes in South America. The aim of the present study was to identify the chemical composition of leaf essential oil of S. quichensis, growing wild in Costa Rica. The extraction of the oils was carried out by the hydrodistillation method, using a modified Clevenger type apparatus. The chemical composition of the oils was analyzed by capillary GC-FID and GC-MS using the retention indices on DB-5 type capillary column. A total of 100 compounds were identified, accounting for about 90 percent of the total amount of the oils. Smallanthus quichensis leaf produced a monoterpenoid-rich oil, whose composition was dominated by αlfa-pinene (64.5 percent) and 1,8-cineole (9.7 percent) or, in a different sample, by αlfa-pinene (35.5 percent) with moderate amounts of p-cymene (11.5 percent), beta-phellandrene (9.2 percent), alfa-phellandrene (9.0 percent) and limonene (5.8 percent). This is the first report of the chemical composition of the essential oil obtained from this plant species.


Smallanthus es un género de plantas perteneciente a la familia Asteraceae que contiene aproximadamente 24 especies, la mayoría ubicadas desde el sur de México, América Central y hasta la cordillera de los Andes en América del Sur. El objetivo del presente estudio fue el de identificar la composición química del aceite esencial de las hojas de S. quichensis. La extracción se realizó mediante el método de hidrodestilación, empleando un instrumento de tipo Clevenger. Se analizó la composición de los aceites mediante CG-FID y CG-EM, utilizando índices de retención obtenidos en una columna capilar tipo DB-5. Se identificaron 100 compuestos, correspondientes a un 90 por ciento de los constituyentes totales. Los aceites están constituidos principalmente por monoterpenoides (ca. 85 por ciento). Los componentes mayoritarios se identificaron como alfa-pineno (64.5 por ciento), acompañado por 1,8-cineol (9.7 por ciento) o, en otra muestra distinta, αlfa-pineno (35.5 por ciento) junto a cantidades moderadas de p-cimeno (11.5 por ciento), beta-felandreno (9.2 por ciento), alfa-felandreno (9.0 por ciento) y limoneno (5.8 por ciento). Este es el primer informe acerca de la composición química de aceites esenciales obtenidos de S. quichensis.

Filmy ferns are a conspicuous epiphytic component of the temperate rain forest of southern Chile and the more abundant genus is Hymenophyllum represented by 19 species, they are a primitive fern family sharing their limited water lost control (poikilohydricity) with lower non-vascular plants, such as bryophytes and lichens. Because, carbohydrate accumulation is a desiccation tolerance mechanism proposed for desiccation-tolerant vascularized plants, the aim of this investigation was to propose a methodology for soluble carbohydrate analysis in native ferns. Preparative- TLC showed to be a simple and rapid technique for separating Hymenophyllum sugars. GC-MS analysis allowed identifying glucitol (2.0 percent), sucrose (2.0 percent), alpha or beta-melibiose (5.0 percent) and salidroside (4.4 percent) in H. cruentum carbohydrate profile from H. dentatum showed significant differences presenting only two sugars sucrose (1.4 percent) and alpha or beta-melibiose (3.2 percent). Furthermore, salidroside, an antioxidant compound, was identified only in H. cruentum. Our results suggest that H. cruentum, has higher preventive mechanisms than H. dentatum; that would allow delay the effects of desiccation (prevent a rapid dehydration) and thus survive to short periods of drought present in the study area. Due to the presence of salidroside in H. cruentum this species should be more resistant to oxidative stress that H. dentatum.


Los helechos película se pueden encontrar en el bosque templado lluvioso del sur de Chile, siendo el género más abundante Hymenophyllum representado con 19 especies. Son una familia de helechos primitivos que comparten la limitación de control de pérdida de agua (poiquilohidrícos) con plantas no vasculares, tales como briófitas y líquenes. Como el contenido de hidratos de carbono ha sido asociado a la protección de la planta durante la desecación, su análisis es una herramienta importante para dilucidar el mecanismo asociado a este fenómeno. El objetivo de esta investigación fue proponer una metodología fácil y rápida para el análisis de hidratos de carbono en helechos nativos. La CCF-preparativa permitió la separación de hidratos de carbono presentes en plantas del género Hymenophyllum. El análisis por GC-MS logró identificar glucitol (2,0 por ciento), sacarosa (2,0 por ciento), alfa o Beta-melibiosa (5.0 por ciento) y salidrosido (4,4 por ciento) en H. cruentum. El perfil de hidratos de carbono de H. dentatum mostró diferencias significativas, mostrando sólo dos compuestos sacarosa (1,4 por ciento) y alfa o beta- melibiosa (3.2 por ciento). Además, salidrosido, un compuesto antioxidante, sólo fue identificado en H. cruentum. Nuestros resultados sugieren que ante la menor tolerancia a la desecación observada previamente en H. cruentum, esta especie presenta mecanismos que le permitirían retardar los efectos de la desecación (prevención de una rápida pérdida de agua) y así sobrevivir a los periodos cortos de sequía presentes en la zona de estudio. Debido a La presencia de salidrosido en H. cruentum, esta especie debería ser más resistente al estrés oxidativo que H. dentatum.

El aceite esencial de Melaleuca quinquenervia de la Ciénaga de Zapata, Cuba, obtenido a escala piloto con un rendimiento promedio de 0,8%, presentó una densidad relativa y un índice de refracción promedios de 0,9253g/mL y 1,4827 respectivamente. La determinación de su composición química por cromatografía de gases acoplada a espectrometría de masas permitió identificar 42 compuestos, de los cuales 26 se sometieron al Análisis por Componentes Principales y al Análisis por Conglomerados. Dichos análisis permitieron la separación de las muestras en dos grupos con diferencias significativas (p<0,05) entre los contenidos promedios de sus principales constituyentes. A partir de dichos análisis se estableció la presencia de dos quimiotipos diferentes: uno con elevados contenidos de viridiflorol (33,8 por ciento), seguido por el 1,8-cineol (21,8 por ciento), y otro quimiotipo del tipo 1,8-cineol (30,0 por ciento) con un contenido relativamente elevado de viridiflorol (24,6 por ciento). Ambos quimiotipos presentaron concentraciones similares de los monoterpenos limoneno y αlfa-pineno.


The essential oil of Melaleuca quinquenervia from Ciénaga de Zapata, Cuba, obtained at pilot scale with average yield of 0.8%, showed average relative density and refraction index of 0.9253g/mL and 1.4827 respectively. It ́s chemical composition determination by gas chromatography coupled to Mass Spectrometry allow identifying 42 compounds, 26 of which were submitted to Principal Components Analysis and to Conglomerates Analysis. Such analyses allowed the separation of the samples into two groups with significant differences (p<0.05) between the average contents of its main components. From such analyses, it was established the presence of two different chemotypes: one with high contents of viridiflorol (33.8 percent), followed by 1,8-cineol (21.8 percent), and another chemotype rich in 1,8-cineol (30,0 percent) with a relatively high content in viridiflorol (24.6 percent). Both chemotypes showed similar concentrations of limonene and αlfa-pinene monoterpenes.

The composition of the essential oil (EO) from Cryptocarya alba obtained by hydro distillation of fresh leaves was analyzed using gas chromatography (GC) and gas chromatography/mass spectrosmetry (GC/MS). The insecticidal effect of the oil on the house fly Musca domestica was evaluated by placing flies in a sealed glass jar containing a piece of EO-treated cotton yarn. The dose necessary to kill 50 percent of flies (LC50) in 0.5 and 1 h was determined at 26 +/- 1 °C. The essential oil from Cryptocarya alba showed significant insecticidal properties [33.56 (7.06-159.50) mg/dm3 (0.5 h) and 15.07 (5.29-42.91) mg/dm3 (1 h)]. According to GC and GC/MS analysis a total of 38 compounds were identified. The composition of the Cryptocarya alba essential oil reported in this study is different to that reported in other publications with 4-terpineol (17.48 percent) ; 4- (3,3-dimethyl-but-1-ynyl) -4-hydroxy-2,6,6-trimethylcyclohex-2-enone (12.84 percent); 1,8-cineole (7.90 percent); p-cymene (7.11 percent) and sabinene (6.80 percent), accounting for 52.13 percent of the EO. The EO from Cryptocarya alba appears promising as a natural insecticide against houseflies.


La composición del aceite esencial (AE), obtenido por hidrodestilación de hojas frescas de Cryptocarya alba se analizó mediante cromatografía de gases (CG) y cromatografía de gases/espectrometría de masas (CG/EM). La actividad insecticida del aceite contra la mosca doméstica, Musca domestica se evaluó colocando las moscas en un frasco de vidrio sellado con un trozo de hilo de algodón tratado con diferentes cantidades de AE. La dosis necesaria para matar el 50 por ciento de las moscas (LC50) en 0,5 y 1 hora se determinó a 26 +/- 1 oC. El aceite esencial de Cryptocarya alba mostró un buen efecto insecticida [33,56 (7,06-159,50) mg/dm3 (0,5 h) and 15,07 (5,29-42,91) mg/dm3 (1 h)]. La composición del aceite esencial de Cryptocarya alba encontrada en este trabajo es diferente al informado en otras publicaciones, con 4-terpineol (17,48 por ciento); 4-(3,3-dimetil-but-1-inil)-4-hidroxi-2,6,6-trimetilciclohex-2-enona (12.84 por ciento); 1,8-cineole (7,90 por ciento); p-cimeno (7,11 por ciento) y sabineno (6,80 por ciento), lo que representa el 52.13 por ciento del AE. El AE de Cryptocarya alba parece prometedor como un insecticida natural contra la mosca doméstica.

ABSTRACT

The profile of volatile compounds of yellow passion fruit juice was analyzed by solid phase microextraction headspace (HS-SPME) and optimized static headspace (S-HS) extraction techniques. Time, temperature, NaCl concentration and sample volume headspace equilibrium parameters was adjusted to the S-HS technique. The gaseous phase in the headspace of samples was collected and injected into a gas chromatograph coupled to a mass spectrometer. In the HS-SPME technique was identified 44 volatile compounds from the yellow passion fruit juice, but with S-HS only 30 compounds were identified. Volatile esters were majority in both techniques, being identified ethyl butanoate, ethyl hexanoate, (3z)-3-hexenyl acetate, hexyl acetate, hexyl butanoate and hexyl hexanoate. Aldehydes and ketones were not identified in S-HS, but were in HS-SPME. β-Pinene, p-cymene, limonene, (Z)-β-ocimene, (E)-β-ocimene, γ-terpinene, α-terpinolene and (E) -4,8-dimethyl-1, 3,7 - nonatriene terpenes were identified in both techniques. This study showed that the S-HS optimized extraction technique was effective to recovery high concentrations of the major volatile characteristics compounds in the passion fruit, such as ethyl butanoate and ethyl hexanoate, which can be advantageous due to the simplicity of the method.


O perfil de compostos voláteis do suco de maracujá-amarelo foi analisado pelas técnicas de extração microextração em fase sólida (SPME-HS) e headspace estático (S-HS). Os parâmetros de equilíbrio do headspace tempo, temperatura, concentração de NaCl e volume da amostra foram ajustados para a técnica S-HS. Em ambas as técnicas de extração, a fase gasosa no headspace das amostras foi recolhida e injetada em cromatógrafo gasoso acoplado a espectrômetro de massas. Na técnica SPME-HS, foram identificados 44 compostosvoláteis no suco do maracujá-amarelo e, em S-HS, foram identificados 30 compostos. Ésteres voláteis foram majoritários em ambas as técnicas, sendo identificados butanoato de etila, hexanoato de etila, (3z)-3-hexenil acetato, hexil acetato, hexil butanoatoe hexil hexanoato. Aldeídos e cetonas não foram identificados com a técnica S-HS. Os terpenos β-pineno, p-cimeno, limoneno, (Z)-β-ocimeno, (E)-β-ocimeno, γ-terpineno, α-terpinoleno e (E)-4,8-dimetil-1,3,7-nonatrieno foram identificados em ambas as tιcnicas.. Este estudo mostrou que a técnica de extração otimizada S-HS foi eficaz para a recuperação de altas concentrações dos principais compostos voláteis característicos do maracujá, como o butanoato de etila e o hexanoate deetila, o que é vantajoso, devido à simplicidade do método.

ABSTRACT

The chemical composition of the essential oil obtained by hydrodistillation from fresh leaves of Blepharocalyx salicifolius (Kunth) O. Berg (Myrtaceae) was analyzed by gas chromatography/mass spectrometry (GC/MS). Twenty-two compounds were identified and a predominance of sesquiterpenes was observed (twenty sesquiterpenes - 96.53 percent). This study presents for the first time the qualitative chemical composition of the essential oil from the leaves of B. salicifolius in their native habitat of Brazilian Cerrado. The qualitative chemical profile of essential oil obtained from the specimen in Cerrado differs from those obtained from specimens collected in biomes of southern parts of South America.


La composición química del aceite esencial obtenido por destilación al vapor de las hojas frescas de Blepharocalyx salicifolius (Kunth) O. Berg (Myrtaceae) se analizó por CG/EM (Cromatografia de Gases/ Espectrometría de Masas). Veintidós compuestos fueron identificados, predominantemente sesquiterpenos (veinte sesquiterpenos - 96.53 por ciento). Este estudio demuestra por primera vez, el análisis químico cualitativo de aceite esencial de hojas de B. salicifolius en su hábitat nativo del Cerrado brasileño. El perfil químico del aceite esencial obtenido de la muestra B. salicifolius del Cerrado difiere de la obtenida a partir de muestras recogidas en los biomas del sur de América del Sur.

ABSTRACT

The piperidine alkaloid composition from young stems of Lobelia polyphylla Hook & Arn. was determined by gas chromatography mass spectrometry (GC-MS). The tentative structures, without the stereochemistry, were obtained by the analysis of the fragmentation patterns of the mass spectra of each compound. The stems contained a mixture of lobeline (1), norlobelanidine (2), 1-(1-(2-hydroxy-2-phenylethyl)-1-methylpiperidin) butane-2-ol (3), 8-propyl-10-phenyl lobelionol (4), 1-(6-(2-hydroxy-2-phenylethyl)-1-methylpiperidin) butane-2-one (5), 1-(6-(2-hidroxypentyl)-1-ethylpiperidin) butane-2-one (6) and 1-methyl-2-piperidinemethanol (7). The role of these alkaloids in the toxic, narcotic and hallucinogenic effects, produced after smoking the aerial parts of this species is discussed.


La composición de alcaloides piperidínicos de tallos jóvenes de Lobelia polyphylla Hook & Arn. se determinó por cromatografía de gases acoplada a espectrometría de masas (CG-EM). Las estructuras tentativas sin incluir la estereoquímica, se obtuvieron mediante el análisis de los patrones de fragmentación de los espectros de masas de cada compuesto. Los tallos contienen una mezcla de lobelina (1), norlobelanidina (2), 1-(1-(2-hidroxi-2-feniletil)-1-metilpiperidin) butano-2-ol (3), 8-propil-10-fenil lobelionol (4), 1-(6-(2-hidroxi-2-feniletil)-1-metilpiperidin) butano-2-ona (5), 1-(6-(2-hidroxypentyl)-1-etilpiperidin) butano-2-ona (6) y 1-metil-2-piperidinmetanol (7). Se discute el posible papel de estos alcaloides en los efectos tóxicos, estupefacientes y alucinógenos, producidos después de haber fumado la parte aérea de esta especie.

ABSTRACT

Aos frutos e óleos essenciais da aroeira da praia (Schinus terebinthifolius) atribui-se atividade antimicrobiana sobre bactérias gram positivas além de ação anti-inflamatória. O óleo essencial da aroeira é utilizado no tratamento de micoses, sendo a sua atividade antifúngica atribuída à alta concentração de monoterpenos. Este estudo teve como objetivo determinar o melhor rendimento de destilação de óleo essencial de sementes e folhas da aroeira e os componentes químicos do óleo essencial nos diversos períodos de destilação. Cem gramas de cada material foram destilados em aparato de Clevenger durante 2,5h, 4,0h, 5,5h e 7,0h. O material destilado foi analisado por cromatografia a gás acoplada a espectrofotômetro de massas, onde foi obtido o perfil cromatográfico do material. As sementes de aroeira apresentaram maiores quantidades de óleo essencial, quando comparadas com as folhas em todos os períodos de destilação. O óleo essencial de aroeira mostrou perfil volátil formado principalmente por mono e sesquiterpenos.


The fruits and essential oils of Brazilian pepper (Schinus terebinthifolius) have antimicrobial activity against gram-positive bacteria, as well as anti-inflammatory action. The essential oil of Brazilian pepper is used to treat fungal infections, and its antifungal activity is attributed to the high concentration of monoterpenes. This study aimed to determine the best yield of distillation of the essential oil of seeds and leaves of Brazilian pepper and the chemical components of the essential oil in different periods of distillation. One hundred grams of each material were distilled in Clevenger apparatus for 2.5 h, 4.0 h, 5.5 h and 7.0 h. The distilled material was analyzed by gas chromatography coupled to mass spectrometer, where the chromatographic profile of the material was obtained. Seeds of Brazilian pepper showed higher amounts of essential oils when compared with the leaves in all the periods of distillation. The essential oil of Brazilian pepper showed a profile consisting mainly of volatile mono- and sesquiterpenes.

ABSTRACT

The present study is pioneer in analyzing the chemical composition of the essential oil from aerial parts of Turnera subulata Sm. and evaluates their antibacterial activity against a panel of drug-resistant strains of Staphylococcus aureus. The aerial parts were kiln-dried and then powdered in mechanical mill, and was subjected to hydrodistillation in a Clevenger-type apparatus. The components were analyzed using a mass spectrometrycoupled gas chromatography (GC-MS), and the identification of substances was performed by comparison of the mass spectra obtained with the mass spectra of the database of the GC-MS and retention indices . The essential oil was evaluated using the method of broth dilution at concentrations ranging from 3.125µg/mL to 3200µg/mL. It was possible to identify 45 substances (92.1%) of the essential oils, and the major components were trans-caryophyllene (6.7%), citronellol (5.6%), sphatulenol (5.3%), α-cadinol (4.3%), n-tricosano (4.3%), geraniol (4.1%) and trans-geranilacetone (3.7%), n-pentacosano (3.5%), globulol (3.4%), caryophyllene oxide (3.2%). The essential oil of T. subulata Sm. showed effective antibacterial activity for the various strains of S. aureus tested, with the MIC values between 25 µg/mL and 1600 µg/mL. The study of the essential oil of Turnera subulata Sm. showed that it consists of a complex mixture of several classes of compounds having the sesquiterpenes as major constituents followed by monoterpenes, and showed an antibacterial activity significant front the strains tested.


O presente estudo é pioneiro em analisar a composição química dos óleos essenciais das partes aéreas de Turnera subulata Sm. e sua atividade antibacteriana frente à Staphylococcus aureus resistentes a antibióticos. As partes aéreas da planta foram secas em estufa, pulverizadas em moinho mecânico e submetidas à hidrodestilaçao em aparato tipo Clevenger. A composição dos óleos essenciais foi analisada por cromatografia gasosa acoplada a espectrometria de massas (CG-EM), e a identificação realizada por comparação dos espectros de massas com a biblioteca do CG-EM e índices de retenção. A atividade antimicrobiana do óleo essencial foi avaliada usando o método de diluição em caldo, em concentrações que variaram de 3.125µg/mL to 3200µg/mL. Foi possível identificar 45 substâncias (92,1%) do óleo essencial, sendo os componentes majoritários: trans-cariofileno (6,7%), citronelol (5,6%), espatulenol (5,3%), α-cadinol (4,3%), n-tricosano (4,3%), geraniol (4,1%) e trans-geranilacetona (3,7%), n-pentacosano (3,5%), globulol (3,4%) e óxido de cariofileno (3,2%). O óleo essencial de T. subulata Sm. mostrou atividade antibacteriana eficaz para as várias cepas de S. aureus testadas, com valores de CIM entre 25 µg/mL e 1600 µg/mL. O estudo do óleo essencial de Turnera subulata Sm. evidenciou sua complexa mistura, contendo várias classes de substâncias, tendo os sesquiterpenos como constituintes majoritários seguido dos monoterpenos, e mostrou significativa atividade antibacteriana frente as cepas testadas.

ABSTRACT

The essential oil obtained by hydrodistillation from dry leaves of Capsicodendron dinisii (Schwancke) was analyzed using gas chromatography (GC) and gas chromatography/mass (GC/MS) spectrometry. According to GC and GC/MS analysis, the essential oil is mostly composed of sesquiterpene hydrocarbons (69.7 percent) and oxygenated sesquiterpenes (22.6 percent). Bicyclogermacrene (30.8 percent), E-caryophyllene (17.2 percent), spathulenol (13.5 percent) and germacrene D (7.6 percent) were the principal components identified in the essential oil.


El aceite esencial obtenido por hidrodestilación de hojas secas de Capsicodendron dinisii (Schwancke) se analizó mediante cromatografía de gases y cromatografía de gases/espectrometría de masas. De acuerdo con los análisis, el aceite esencial contiene un 69,7 por ciento de sesquiterpenos hidrocarbonados y 22,6 por ciento de sesquiterpenos oxigenados. Bicyclogermacreno (30.8 por ciento), E-cariofileno (17.2 por ciento), espathulenol (13.5 por ciento) and germacreno D (7.6 por ciento) fueron los componentes principales identificados en el aceite esencial.

ABSTRACT

Three phenolic aristolactams, aristolactam AII (3), velutinam (4) and piperolactam A (5), were identified from the leaves and stems of Aristolochia chilensis Bridges ex Lindl. The structures of these compounds were elucidated using a combination of HPLC-DAD, GC-MS and NMR experiments.


Tres aristolactamas fenólicas aristolactama AII(3), velutinam(4) y piperolactama A(5), se identificaron en hojas y tallos de Aristolochia chilensis Bridges ex Lindl. Las estructuras de estos compuestos se determinaron por combinación de CLAE-DAD, CG-EM y experimentos de RMN.

ABSTRACT

Introducción: el D-003 es un ingrediente farmacéutico activo purificado a partir de la cera de caña de azúcar (Saccharum officinarum L.) con efectos como reductor del colesterol y antioxidante, el cual está compuesto por una mezcla de ácidos grasos libres saturados de elevado peso molecular, cada uno dentro de un intervalo de concentración específica determinada por cromatografía de gases (CG). La caracterización espectroscópica del D-003, sin embargo, no ha sido previamente informada. Objetivo: caracterizar el ingrediente farmacéutico activo nombrado D-003 de acuerdo con sus espectros ultravioleta (UV), infrarrojo (FTIR), resonancia magnética nuclear (RMN) y de masas (EM). Métodos: se evaluaron muestras de seis lotes de D-003 (CNIC, Cuba) mediante las técnicas de UV, FTIR, RMN-¹H, RMN-13C y CG-EM. Para obtener los espectros de masas de los ácidos del D-003, estos se derivaron como ésteres metílicos y trimetilsilil. La cuantificación de los ácidos grasos libres saturados de elevado peso molecular, analizados como ésteres metílicos, se llevó a cabo por CG con detector de ionización por llama (DILL). Resultados: los espectros UV, IR, RMN-¹H y RMN-13C mostraron que el ingrediente farmacéutico activo D00-3 está constituido por una mezcla de ácidos grasos libres saturados de elevado peso molecular, mientras las técnicas de CG-EM y CG-DILL permitieron asegurar la presencia de 13 ácidos grasos libres saturados de elevado peso molecular en proporciones definidas: C24:0 (0,2-0,6 por ciento), C25:0 (0,4-0,9 por ciento), C26:0 (2,0-3,1 por ciento), C27:0 (2,1-2,7 por ciento), C28:0 (30,0-37,5 por ciento), C29:0 (1,5-1,7 por ciento), C30:0 (17,0-18,6 por ciento), C31:0 (0,9-1,2 por ciento), C32:0 (6,9-8,9 por ciento), C33:0 (0,9-1,3 por ciento), C34:0 (7,2-11,1 por ciento), C35:0 (0,3-0,6 por ciento) y C36:0 (2,2-3,8 por ciento)(AU)


Introduction: D-003, an active pharmaceutical ingredient (API) purified from sugar cane (Saccharum officinarum L.) wax with cholesterol-lowering and antioxidant effects, is composed of a mixture of free saturated very long chain fatty acids (VLCFAs), each within specific relative concentration ranges as determined by the gas chromatography (GC). However, the spectroscopic characterization of D-003 had not been previously reported. Objective: to characterize the active pharmaceutical ingredient named D003 in accordance to its ultraviolet (UV), infrared (FTIR), nuclear magnetic resonance (NMR) and mass (MS) spectra. Methods: samples of six batches of D-003 (CNIC, Cuba) were evaluated by UV, FTIR, NMR-¹H, NMR-13C, and GC-MS techniques. For obtaining the mass spectra of D-003 acids, methyl (FAME) and trimethylsilyl ester derivatives were used. Quantification of free very long chain saturated fatty acids , analyzed as methyl ester derivatives, was made by GC with flame ionisation detector (FID). Results: UV, FTIR, and ¹H-NMR, 13C-NMR spectra showed that active pharmaceutical ingredient D-003 was composed of a mixture of free very long chain fatty acids, whereas the GC-MS and GC-FID techniques allowed ensuring the occurrence of 13 VLCFAs in set proportions: C24:0 (0.2-0.6 percent), C25:0 (0.4-0.9 percent), C26:0 (2.0-3.1 percent), C27:0 (2.1-2.7 percent), C28:0 (30.0-37.5 percent), C29:0 (1.5-1.7 percent), C30:0 (17.0-18.6 percent), C31:0 (0.9-1.2 percent), C32:0 (6.9-8.9 percent), C33:0 (0.9-1.3 percent), C34:0 (7.2-11.1 percent), C35:0 (0.3-0.6 percent) and C36:0 (2.2-3.8 percent). Conclusions: evidences from the UV, FTIR, NMR and GC-MS spectroscopic techniques prove that D-003 is composed of 13 saturated very long chain fatty acids, including octacosanoic acid as the most abundant one(AU)

ABSTRACT

Cotinine is the major metabolite of nicotine and, being very stable and having a long biological half-life, it can be used as a biomarker for tobacco exposure. The aim of this study was to develop an analytical GC-MS technique to measure levels of cotinine in the urine of active and passive smokers and to compare the results with reference values. The extraction of cotinine to generate the calibration curve was performed by mixing urine (250 ?L) with 50 ?L of a cotinine standard, 50 ?L of an internal standard of deuterated cotinine (15μgμmL-1) and 50 μL of 10% NH4OH solution. Next, 2 mL of a mixture of MTBE:dichloromethane:ethyl acetate (30:30:40 by volume) was added and the whole was vortexed, then centrifuged at 3000 rpm. Finally, 1.6 mL of the organic layer was evaporated under a stream of dry air at 50 °C. The resulting extract was dissolved in methanol and injected into the GC-MS system. The LOQ and LOD for cotinine were 100 and 20 ng.mL-1, respectively. The curve was linear over the whole tested range of 100 - 5000 ng.mL-1 and the method achieved 50% recovery. The intra and inter-day precisions were 1.62 ? 7.28% and 0.86 - 2.68%, respectively. Accuracy was determined at three concentrations (low, medium and high), with six replicates (95.24- 97.67%). The validation of this cotinine assay by GC-MS showed that it exhibited satisfactory limits and the assay could be performed with a one-step liquid-liquid extraction. The technique presented here can thus be used for the quantitation of cotinine levels in the urine of passive and active smokers.


A cotinina é o metabólito principal da nicotina, é muito estável e tem uma elevada meia-vida biológica e pode ser usado como biomarcador da exposição ao tabaco. O objetivo deste estudo foi desenvolver uma técnica analítica em CG-EM para medir os níveis de cotinina na urina de fumantes ativos e passivos e comparar os resultados com valores de referência. A extração da cotinina para construir a curva de calibração foi desenvolvida misturando 250 μL de padrão de cotinina em urina, 50 μL de padrão interno (cotinina deuterada 15 ?g?mL-1) e 50 μL de solução aquosa de NH4OH 10%. Em seguida, 2 mL da mistura MTBE:diclorometano:acetato de etila (30:30:40 v/v) foi adicionada, agitada em vórtex e centrifugada a 3000 rpm. Finalmente, 1,6 mL da camada orgânica foi evaporada sob ar seco a 50 °C. O extrato resultante foi dissolvido em metanol e injetado no sistema CG-EM. Os limites de quantificação e de detecção foram 100 e 20 ng?mL-1, respectivamente. A curva de calibração foi linear no intervalo de concentração testado (100 - 5000 ng?mL-1), com 50% de recuperação. Os valores de precisão intra-dia e inter-dia foram 1,62 ? 7,28% e 0,86 - 2,68%, respectivamente. A exatidão (95,24 - 97,67%) foi determinada sob 3 concentrações (baixa, média e alta), com 6 replicatas. A validação deste procedimento para análise de cotinina por CG-EM demonstrou valores satisfatórios, numa única etapa de extração líquido-líquido, podendo ser utilizada para a quantificação dos níveis de cotinina em amostras de urina de fumantes ativos e passivos.

Introducción: los esteroles han sido ampliamente estudiados por su importancia nutricional y farmacéutica. Sin embargo, para nuestro conocimiento, la composición de esteroles del aceite de los frutos de la palma real cubana (Roystonea regia) no ha sido determinada hasta el momento. Objetivos: determinar la composición de esteroles que pudieran estar presentes en la fracción insaponificable del aceite de R. regia mediante CG-EM. Métodos: muestras de dos lotes de aceite de R. regia fueron sometidas al procedimiento para la determinación de esteroles establecido por el del Instituto de Nutracéuticos de los EE.UU., el cual consiste, fundamentalmente, en una saponificación con disolución de KOH/EtOH y posterior extracción con n-hexano de las fracciones insaponificables. Las fracciones obtenidas fueron analizadas por CG-EM como derivados TMS e identificadas por comparación de sus espectros con los de patrones comerciales y los de la base de espectros Wiley. Para la cuantificación se utilizó el colestano como patrón interno. Resultados: en la fracción insaponificable del aceite de R. regia se encontró un contenido total de esteroles de 66,1por ciento; lo que representa un 0,14 por ciento del aceite. La fracción de esteroles, estuvo compuesta principalmente por â -sitosterol (51,2 por ciento), estigmasterol (9,6 porciento), campesterol (9,2 por ciento), 24-metilen-cicloartanol (9,2 porciento), Ã5-avenasterol (8,9 por ciento), cicloartanol (7,5 por ciento); además de otros componentes minoritarios como cicloartenol, ã-sitosterol y colesterol. Conclusiones: se identificaron y se cuantificaron mediante CG-EM los esteroles de la fracción insaponificable del aceite de R. regia, en la cual el â-sitosterol resultó el componente mayoritario. Estos resultados son una contribución al estudio de la composición química de dicho aceite y pudieran avalar su posible utilidad nutricional y seguridad


Introduction: sterols have been widely studied because of their pharmaceutical and nutritional importance. However, to our knowledge, the sterol composition of the oil from the Cuban Royal Palm fruits (Roystonea regia (Kunth) F. Cook), has not been yet identified Objectives: to determine by GC-MS the sterols that could be present in the unsaponifiable fraction of R. regia oil. Methods: samples of two batches of R. regia oil were subjected to the established procedure of the US Institute for Nutraceutical Advancement to determine sterols, which mainly consists of saponification with KOH/EtOH solution and a subsequent extraction of the unsaponifiable fractions by using n-hexane. These fractions were analyzed by GC-MS as TMS derivatives and they were identified by comparing their spectra with those of commercial available standards and with spectra of the Wiley mass spectrum library. Quantification was made by using cholestane as internal standard. Results: a total sterol content of 66.1percent was found in the unsaponifiable fraction of R. regia oil, which represents 0.14 percent from the oil. The sterol fraction was mainly composed of â-sitosterol (51.2 percent), stigmasterol (9.6 percent), campesterol (9.2 percent), 24-methylen-cycloartanol (9.2 percent), Ã5-avenasterol (8.9 percent) and cycloartanol (7.5 percent) in addition to other minor components such as cycloartenol, ?-sitosterol, and cholesterol. Conclusions: sterol compounds were identified and quantified by GC-MS in the unsaponifiable fraction of R. regia oil, in which the â-sitosterol was the predominant component. These results are a contribution to the chemical composition study of such oil and could support its possible nutritional usefulness and safety

ABSTRACT

The analysis by gas chromatography coupled with mass spectrometry (GC-MS) of the alkaloidal extract of Browningia candelaris (Cactaceae) showed the presence of N-acetyl-3,4-dimethoxyphenylethylamine; N,N-dimethyl-3,4-dimethoxyphenylethylamine; N,N-dimethyl-4-methoxyphenylethylamine; and 4-methoxyamphetamine. The presence of these psychoactive compounds is discussed in terms of their possible ritual use in Andean cultures of Northern Chile.


El análisis por medio de cromatografía de gases acoplada a espectrometría de masas (CG-EM) del extracto alcaloidal de Browningia candelaris (Cactaceae) mostró la presencia de N-acetil-3,4-dimetoxifeniletilamina; N,N-dimetil-3,4-dimetoxifeniletilamina; N,N-dimetil-4-metoxifeniletilamina y 4-metoxianfetamina. La presencia de estos compuestos psicoactivos se discute en términos de su posible utilización en ceremonias mágico-religiosas por culturas andinas del norte de Chile.
